Semaglutide GLP-1 receptor agonist peptide: a research overview
Few peptides have been studied as thoroughly as semaglutide. Here's a cozy summary of the major published trials and where things stand.

Published findings
Semaglutide's evidence base rests on large randomized phase 3 trials. In the STEP 1 trial (NEJM, 2021), adults with overweight or obesity who also followed a lifestyle program saw an average body-weight change of −14.9% by week 68, versus −2.4% on placebo [1]. The SELECT trial (NEJM, 2023) followed 17,604 adults who had cardiovascular disease and overweight or obesity without diabetes for a mean of 39.8 months; major cardiovascular events occurred in 6.5% of the semaglutide group and 8.0% of the placebo group (hazard ratio 0.80), while stopping treatment because of adverse events was more common on semaglutide (16.6% vs 8.2%) [2].
Mechanism at a glance
At the receptor level, semaglutide stands in for glucagon-like peptide-1, switching on that hormone's receptor [3]. Its labeled uses include glycemic control in type 2 diabetes [4].
Regulatory & WADA notes
Semaglutide is the active ingredient in two FDA-approved medicines. Ozempic is indicated for glycemic control in adults with type 2 diabetes and for reducing certain cardiovascular and kidney risks in that population [4]. Wegovy is indicated for long-term weight reduction, for cardiovascular risk reduction in adults with established cardiovascular disease and obesity or overweight, and for noncirrhotic MASH with moderate-to-advanced fibrosis [3]. This research material is not one of those products. In sport, semaglutide does not appear on WADA's 2026 Prohibited List; WADA's 2026 Monitoring Program tracks markers of semaglutide and tirzepatide [5].
